Knudson's two-hit hypothesis of tumorigenesis

Knudson’s two-hit hypothesis of tumorigenesis claims that a germline mutation of a tumor suppressor gene is accompanied by an additional somatic mutation. Then the two alleles of a protective gene being silenced promotes tumorigenesis.
