Apolipoprotein A5
Das APOA5 Gen kodiert ein Lipoprotein, welches ein wichtiger Bestandteil der HDL-Partikel ist. Polymorphismen sind mit hohen Triglyceridspiegeln, niedrigen HDL-Werten und der koronaren Herzkrankheit assoziiert. Loss-of-function Mutationen können eine schwere Hypertriglyceridämie und gelegentlich sogar eine Chylomikronämie hervorrufen, insbesondere wenn homozygot.
Interpretation
Loss-of-function Mutationen des APOA5-Gens sind selten. Sie können bereits im heterozygoten Zustand zu einer deutlichen Hypertriglyceridämie führen. Im homozygoten oder compound heterozygoten Zustand kann es zur Chylomikronämie kommen. Auch ließen sich verschiedene häufigere Polymorphismen mit einer Neigung zu erhöhten Triglyceridspiegeln und kardiovaskulären Erkrankungen assoziieren.
