Das Nagel-Patella-Syndrom ist eine autosomal dominante Erkrankung. Nagelveränderungen, sowie das Fehlen oder die Hypoplasie der Patella sind die hervorstechendsten Symptome. Weiterhin können auch noch andere Skelettveränderungen und eine hereditäre Nephritis vorkommen.
Die Häufigkeit der Erkrankung liegt bei etwa 1:50.000 Lebendgeborenen. Mit dem Alter des Vaters scheint die Wahrscheinlichkeit für Neumutationen zu steigen.
Am hervorstechendsten sind die Skelettveränderungen bei der Onychoosteodysplasie. Dazu gehören eine Hypoplasie oder Aplasie der Patella, Dystrophie der Ellenbogen, Nagelveränderungen (insbesondere am Daumen) und Knochenwülste an den Beckenschaufeln (sogenannte Beckenhörner). Weiterhin tritt eine Nephropathie auf und Glaukome sind bei diesen Patienten häufiger.
Patienten mit Nagel-Patella-Syndrom sind in ihrer geistigen und körperlichen Leistungsfähigkeit wenig beeinträchtig. Die Lebenserwartung ist normal.
Die Onychoosteodysplasie wird durch Mutationen des LMX1B-Gens ausgelöst.
